Investigational Factor Xa Compound: Data Presented At European Heart Meeting
While the results of the study did non show a statistically important difference in ischemic events among any of the four doses of apixaban evaluated, trends emerged that support farther study, the scientists said.
Apixaban, one of several new anti-platelet agents under exploitation, targets the activity of Factor Xa, an enzyme involved in the process of rake coagulation. Physicians say the need for safer, better anticoagulants is critical because current therapies fall short of therapeutical goals or are difficult to pull off because they increase the risk of bleeding.
"One of the nearly vexing problems in cardiology is identifying the correct combination of drugs that can subdue clot formation but non increase the risk of serious hemorrhage," said study lead John Alexander, M.D., a heart specialist at Duke University Medical Center, world Health Organization presented the findings at the European Society of Cardiology meeting in Munich.
Millions of patients worldwide take anti-clotting drugs on a regular ground. Most are prescribed empirin, clopidogrel (likewise known as Plavix or Iscover) and/or warfarin.
"Warfarin is clearly effective as an anticoagulant, only it is especially hard to pull off properly and safely," Alexander said. "What's driving research in this area is the fact that although aspirin and clopidogrel both offer spunk attack patients important auspices against blood clots, there is still room for improvement. Studies show that a significant number of patients taking aspirin and clopidogrel volition still have some sort of job related to a blood clot. Adding warfarin just increases the likelihood of bleeding complications at a higher rate than anybody is comfortable with."
Alexander, along with Professor Lars Wallentin from the Uppsala Clinical Research Center in Sweden and other colleagues, studied the use of goods and services of apixaban in 1,715 patients from 14 countries end-to-end Europe and North America who had suffered a recent spirit attack. Roughly two-thirds of the patients had undergone angioplasty to clear plugged arteries and 99 pct of them were pickings either empirin or acetylsalicylic acid and clopidogrel.
The study (called APPRAISE) was designed to identify the optimal dose of apixaban. Participants were randomized into one of four doses of apixaban or a placebo. Researchers tracked the incidence of bleeding and recurrent kernel attack, accident, chest pain requiring hospitalization or additional procedures or death from cardiovascular problems for the following six-spot months.
At the end of the field, researchers discovered a non-statistically significant movement suggesting that patients taking apixaban along with their regular treatment had a lower incidence of heart attack, stroke, chest pain or decease from cardiovascular problems than patients pickings a placebo.
But bleeding was an issue, especially among those taking the higher doses of the drug (10 milligrams twice daily or 20 milligrams erst daily). Investigators discontinued those two blazonry of the study because patients were experiencing unsufferable rates of bleeding. Patients in the remaining munition of the study (2.5 milligrams twice daily or 10 milligrams once daily) also experienced more bleeding than those pickings a placebo.
Investigators say that while the results ar not conclusive, they do warrant further study.
"The information show adding 5 or 10 milligrams of apixaban to a regimen of aspirin or aspirin and clopidogrel in patients hoping to keep a s heart attack may offer therapeutic potency," says Alexander. "But this needs to be definitively demonstrated in a statistically significant mode in magnanimous, well-controlled studies."
Apixaban is under development at Bristol-Myers Squibb, the company that sponsored the study.
Alexander and Wallentin have both received inquiry support from Bristol-Myers Squibb.

Iva Davies goes into Battle
You've turned down reality TV in the past, but now you're judging on Battle of the Choirs. What made you say yes? This appeals to me because I don't think the people went into their choirs to be TV stars or music stars or dancing stars. They just love doing what they're doing and they'd do it whether there was a TV show there or not. Reality-TV shows love tension between the judges. How's the vibe on Choirs? There have been some tense moments when the judges have been divided. I don't watch any reality TV, I'm a sucker for a good documentary. Your fellow judge Charli from Hi-5 said she was the only one who knew how Pink's Get the Party Started should be sung. True? I'm vaguely aware of that stuff, I have children who are 14 and 12. It's not that I study the charts any more, but a lot of those songs have filtered through. That Mika song (Grace Kelly) was one of my son's favourites for months. Do you listen to much new music? I was a devotee of Pink Floyd and more recently Radiohead, but I've disappeared into that wonderful middle age of life where none of these things become a passion to follow. I think that's the case with many professional musicians, what's recreation to other people is work to us.

Cannes Film Festival - Movie Reviews Changeling
Clint Eastwood's Changeling
has drawn some of the strongest early reviews yet at the Cannes Film Festival. (Critics
were unsure what the actual title was. The festival said on Tuesday that it had received
a written notice from the studio that the title had been changed to The Exchange
, aligning it with the French title, L'Échange. But at a news conference
Eastwood implied that he had not agreed to a such a change, and the daily festival
program, after revising the title on Tuesday changed it back to The Changeling today.)
Geoffrey MacNab in the London Independent called the film a "magisterial piece
of work." While predicting that the movie won't become a big box-office success for
Eastwood, he concluded that "in its unfussy craftsmanship and emotional punch, it shows
him still at the peak of his powers." Writing for the British trade publication Screen
Daily, Mike Goodridge predicted that the movie will "go all the way from the
Palais to the Academy Awards." Several critics attending the screening came away forecasting
an Oscar nomination not only for Eastwood but also for the film's star, Angelina
Jolie. "She is the entire, anxious point of Eastwood's film," wrote James Christopher
in the London Times, "and absolutely terrific." Todd McCarthy in Daily Variety
called her performance "top notch." And Kurt Honeycutt wrote in the Hollywood
Reporter, "Jolie puts on a powerful emotional display."

Rivers Cuomo - Alone: The Home Recordings of Rivers Cuomo
Rivers Cuomo - Alone: The Home Recordings of Rivers Cuomo
In an eRA where the majority of rock candy stars aren't very interesting, there's often to like about the kookiness of Rivers Cuomo.
Having apparently lived in a wardrobe for weeks on end; sold all his possessions browning automatic rifle a guitar to buy a one-room basement flat; left hand tilt 'n' hustle for a level at Harvard University, and gone on sex binges followed by age of abstinence, Weezer's frontman has, justifiedly or wrongly, garnered a report as rock's weirdie.
Something of a hesitant rock star, it is peculiar then that Cuomo has straightaway decided to raid his nigh 20-year appeal of demo recordings and allowed us in.
Recorded between 1992 and 2007, the 18-track accumulation is to a greater extent a peculiarity for Weezer fans than anything else with the quality of a lot of the recordings questionable. With many, it's soft to see why they haven't made it on to Weezer albums, patch others point to Cuomo's sometime desire to experiment outside of Weezer's structured sound and prickteaser with other genres.
'This Is The Way', single of the more recent demos, was seemingly considered as a lead 1 for the band's forthcoming album and is ace of the to a greater extent experimental sounding tracks here in that it has a sleazy hip-hop palpate to it. You can characterisation bling bling, gold and inkiness rappers with Cuomo looking odd and out of place somewhere in-between.
Cuomo's geek-eyed sensitivity flows to the magnetic core on tracks like 'Wanda (You're My Only Passion)', which was intended to soundtrack the motion-picture show 'Angus', and the excellent 'Chess' and 'Longtime Sunshine'.
'Blast Off', however, is the standout runway and i which would feel at nursing home on any Weezer record album.
Recorded as part of the band's second album roger Sessions, Cuomo had intended to release a "space-voyage rock opera" entitled 'Songs from the Blackhole', and so many of the songs here were intended for that influence.
Hardcore Weezer fans will be companion with these tracks, along with much of 'Alone'. Many of the songs get antecedently cropped up online, along with a legion of other to a lesser extent focussed demos.
I of the best things around the record, notwithstanding, is Cuomo's arm notes. The songster openly describes how for each one track came around and what was leaving on in his life sentence at the time.
It's an engaging companion to the 18-tracks and, thankfully, 'Alone' doesn't look like organism the only road into the workings of Cuomo's creative judgement. He's presently looking on a publishing house for his memoirs - now that should prove fascinating.
